    It\u27s all in the lens: Differences in views on obesity prevention between advocates and policy makers

    Background: Intervention strategies to reduce obesity include policy and environmental changes that are designed to provide opportunities, support, and cues to help people develop healthier behaviors. Policy changes at the state level are one way to influence access, social norms, and opportunities for better nutrition and increased physical activity among the population. Methods: Ten states were selected for a broad variance in obesity rates and number of enacted obesity prevention policies during the years of 2006–2009. Within the selected states, a purely qualitative study of attitudes of childhood obesity policy using semistructured telephone interviews was conducted. Interviews were conducted with state policy makers who serve on public health committees. A set of six states that had more than eight childhood obesity policies enacted were selected for subsequent qualitative interviews with a convenience sample of well-established advocates. Results: Policy makers in states where there was more childhood obesity policy action believed in the evidence behind obesity policy proposals. Policy makers also varied in the perception of obesity as a constituent priority. The major differences between advocates and policy makers included a disconnect in information dissemination, opposition, and effectiveness of these policies. Conclusions: The findings from this study show differences in perceptions among policy makers in states with a greater number of obesity prevention bills enacted. There are differences among policy makers and advocates regarding the role and effectiveness of state policy on obesity prevention. This presents an opportunity for researchers and practitioners to improve communication and translation of evidence to policy makers, particularly in states with low legislation

    Fruit distribution is pivotal in shaping the future of both agriculture and agricultural robotics, paving the way for a streamlined supply chain. This study introduces an innovative methodology that harnesses the synergy of RGB imagery, LiDAR, and IMU data, to achieve intricate tree reconstructions and the pinpoint localization of fruits. Such integration not only offers insights into the fruit distribution, which enhances the precision of guidance for agricultural robotics and automation systems, but also sets the stage for simulating synthetic fruit patterns across varied tree architectures. To validate this approach, experiments have been carried out in both a controlled environment and an actual peach orchard.     The Indirect Search for Dark Matter with IceCube

    We revisit the prospects for IceCube and similar kilometer-scale telescopes to detect neutrinos produced by the annihilation of weakly interacting massive dark matter particles (WIMPs) in the Sun. We emphasize that the astrophysics of the problem is understood; models can be observed or, alternatively, ruled out. In searching for a WIMP with spin-independent interactions with ordinary matter, IceCube is only competitive with direct detection experiments if the WIMP mass is sufficiently large. For spin-dependent interactions IceCube already has improved the best limits on spin-dependent WIMP cross sections by two orders of magnitude. This is largely due to the fact that models with significant spin-dependent couplings to protons are the least constrained and, at the same time, the most promising because of the efficient capture of WIMPs in the Sun. We identify models where dark matter particles are beyond the reach of any planned direct detection experiments while being within reach of neutrino telescopes.     An ultralong CDRH2 in HCV neutralizing antibody demonstrates structural plasticity of antibodies against E2 glycoprotein

    A vaccine protective against diverse HCV variants is needed to control the HCV epidemic. Structures of E2 complexes with front layer-specific broadly neutralizing antibodies (bNAbs) isolated from HCV-infected individuals, revealed a disulfide bond-containing CDRH3 that adopts straight (individuals who clear infection) or bent (individuals with chronic infection) conformation. To investigate whether a straight versus bent disulfide bond-containing CDRH3 is specific to particular HCV-infected individuals, we solved a crystal structure of the HCV E2 ectodomain in complex with AR3X, a bNAb with an unusually long CDRH2 that was isolated from the chronically-infected individual from whom the bent CDRH3 bNAbs were derived. The structure revealed that AR3X utilizes both its ultralong CDRH2 and a disulfide motif-containing straight CDRH3 to recognize the E2 front layer. These results demonstrate that both the straight and bent CDRH3 classes of HCV bNAb can be elicited in a single individual, revealing a structural plasticity of VH1-69-derived bNAbs

    The Genomic Distribution and Local Context of Coincident SNPs in Human and Chimpanzee

    We have previously shown that there is an excess of sites that are polymorphic at orthologous positions in humans and chimpanzees and that this is most likely due to cryptic variation in the mutation rate. We showed that this might be a consequence of complex context effects since we found significant heterogeneity in triplet frequencies around coincident single nucleotide polymorphism (SNP) sites. Here, we show that the heterogeneity in triplet frequencies is not specifically associated with coincident SNPs but is instead driven by base composition bias around CpG dinucleotides. As a result, we suggest that cryptic variation in the mutation rate is truly cryptic, in the sense that the mutation rate does not appear to depend on any specific primary sequence context. Furthermore, we propose that the patterns around CpG dinucleotides are driven by the mutability of CpG dinucleotides in different DNA contexts. We also show that the genomic distribution of coincident SNPs is nonuniform and that there are some subtle differences between the distributions of single and coincident SNPs. Furthermore, we identify regions that contain high numbers of coincident SNPs and suggest that one in particular, a region containing the gene PRIM2, may be under balancing selection

    Applying efficient implicit nongeometric constraints in alchemical free energy simulations

    Several strategies have been developed for satisfying bond lengths, angle, and other geometric constraints in molecular dynamics simulations. Advanced variations of alchemical free energy perturbation simulations, however, also require nongeometric constraints. In our recently developed multisite λ‐dynamics simulation method, the conventional λ parameters that are associated with the progress variables in alchemical transformations are treated as dynamic variables and are constrained such that: 0 ≤ λ i ≤ 1 and ∑   i = 1 N λ i = 1. Here, we present four functional forms of λ that implicitly satisfy these nongeometric constraints, whose values and forces are facile to compute and that yield stable simulations using a 2 fs integration timestep. Using model systems, we present the sampling characteristics of these functional forms and demonstrate the enhanced sampling profiles and improved convergence rates that are achieved by the functional form: lambdai=ecsinthetaioversumnolimitsj=1Necsinthetajlambda _i = {{e^{csin theta _i } } over {sumnolimits_{j = 1}^N {e^{csin theta _j } } }} that oscillates between λ i = 0 and λ i = 1 and has relatively steep transitions between these endpoints. © 2011 Wiley Periodicals, Inc.     Concurrent Silicosis and Pulmonary Mycosis at Death

    To examine risk for mycosis among persons with silicosis, we examined US mortality data for 1979–2004. Persons with silicosis were more likely to die with pulmonary mycosis than were those without pneumoconiosis or those with more common pneumoconioses. Health professionals should consider enhanced risk for mycosis for silica-exposed patients
